Cannabis is the most widely abused illicit drug in the US. Results from the 2013 National Survey on Drug Use and Health show that between 2007 and 2012 the rate of marijuana use rose from 5.8% to 7.3%. As use of marijuana increases, it becomes more prevalent in clinical and forensic case work, particularly in impaired driving cases. In 2007 the National Highway Traffic Safety Administration (NHTSA) reported that 8.6% of nighttime drivers tested positive for cannabinoids—a rate almost four times higher than those with blood alcohol levels equal to or above 0.8 g/L. Recent marijuana use is associated with a 2–6 fold increased crash risk, depending on dose, than when unimpaired.   The LC-MS/MS method developed in this study minimizes required sample volume and provides sensitive quantitation of THC, 11-OH-THC and THC-COOH in whol...

Automation Compatibility: Hamilton and Integra Oral fluid is gaining popularity as an alternative matrix for drug analysis. Oral fluid collection is simple, non-invasive, and can be easily monitored. Like other complex biological matrices, sample preparation is required to remove matrix interferents from oral fluid samples prior to LC-MS/MS analysis. This procedure can be time-consuming and is often the “bottle neck” for laboratory analysis and throughput. Dispersive Pipette XTRaction methods require much less solvent and time-consuming steps compared to other SPE techniques. Automation of the extraction is easy to achieve with XTR tips for Hamilton or Integra liquid handlers. Automated Dispersive Pipette XTRaction is highly reproducible and provides the sensitivity necessary for forensic and clinical purposes.   1 Condition Using Mix...

Cleanup Method for Rapid Analysis in Biological Samples Benzodiazepines and sedatives are some of the most commonly abused drugs in the nation and are often found in cases involving driving under the influence of drugs, sexual assaults and overdoses. Unfortunately, most existing methods used to identify benzodiazepines and sedatives across matrices are labor intensive and time-consuming. A simple cleanup method that can be applied across multiple biological matrices would be an ideal solution to increase the throughput capabilities of forensic laboratories. Unlike traditional methods for solid phase extraction, dispersive pipette extraction technology for clean-up focuses on removing matrix interferences, while keeping the benzodiazepines in the sample solution. XTR tips containing loose weak anion exchange (WAX) sorbent remove phospholipids and small non-amphoteric acids...

Extraction < 15 minutesAutomation Compatibility: Hamilton The need for fast robust methods of evaluating cannabinoid content continues to rise with the increase in cannabis use. The chemical complexity of cannabis plant material and subsequent extracts may benefit from solid phase extraction prior to LC-MS/MS analysis. Current procedures either have very laborious sample preparation methods or lack them altogether. An analytical procedure for the determination of 9 cannabinoids in plant material was developed using Dispersive Pipette XTRaction on a Hamilton Microlab NIMBUS96 platform followed by LC-MS/MS analysis. Dispersive Pipette XTRaction requires minimal sorbent resulting in reduced matrix effects and reasonable levels of THC to avoid saturating the detector. The automated extraction method can process up to 96 samples simultaneously in u...
